چکیده

We constructed an optical system and developed the software to create and analyze the fluorescence anisotropy images of a variety of objects including single molecules, quantum dots, proteins, nucleic acids and cells. The system includes an inverted microscope, three calcite prisms (CPs) inserted in the excitation and emission optical paths, and CCD camera. The system allows to record dynamic changes in fluorescence anisotropy images with a millisecond resolution. The interactive program in MatLab has been developed to construct and analyze the fluorescence anisotropy images. Using this technique we analyzed fluorescent properties of thousands of single QDs attached to a glass surface or to actin filaments (QD-nanowires). Despite the “blinking effect” the anisotropy of fluorescence of single QD remains stable and depends mostly on the 3-dimensional orientation of the QD. The fluorescence anisotropy images of QD-actin nanowires demonstrate the variation of anisotropy along and across filaments. The same system was used to study actin filament dynamics in live cells expressing actin fused with green fluorescence protein (GFP).

Imaging molecular interactions in cells by dynamic and static fluorescence anisotropy (rFLIM and emFRET).

We report the implementation and exploitation of fluorescence polarization measurements, in the form of anisotropy fluorescence lifetime imaging microscopy (rFLIM) and energy migration Förster resonance energy transfer (emFRET) modalities, for wide-field, confocal laser-scanning microscopy and flow cytometry of cells. These methods permit the assessment of rotational motion, association and pro...

A Thin Layer Imaging with the Total Internal Reflection Fluorescence Microscopy

Total internal reflection fluorescence microscopy (TIRFM) is an optical technique that allows imaging of a thin layer of the sample with a thickness of about 100-200 nm. It is used in science of cell biology to study cellular processes, especially near the membranes of living cells. This method is based on the total internal reflection phenomenon, where the evanescent wave is generated in the l...

Steady-State Acceptor Fluorescence Anisotropy Imaging under Evanescent Excitation for Visualisation of FRET at the Plasma Membrane

We present a novel imaging system combining total internal reflection fluorescence (TIRF) microscopy with measurement of steady-state acceptor fluorescence anisotropy in order to perform live cell Förster Resonance Energy Transfer (FRET) imaging at the plasma membrane. We compare directly the imaging performance of fluorescence anisotropy resolved TIRF with epifluorescence illumination. The use...

Measuring F-actin properties in dendritic spines

During the last decade, numerous studies have demonstrated that the actin cytoskeleton plays a pivotal role in the control of dendritic spine shape. Synaptic stimulation rapidly changes the actin dynamics and many actin regulators have been shown to play roles in neuron functionality. Accordingly, defects in the regulation of the actin cytoskeleton in neurons have been implicated in memory diso...

Fluorescence imaging of membrane dynamics in living cells.

Methods of wide-field fluorescence microscopy for measuring membrane dynamics of living cells are described, including spectral imaging as well as anisotropy imaging of the membrane marker 6-dodecanoyl-2-dimethylamino naphthalene (laurdan). Plasma membranes are selected by illumination with an evanescent electromagnetic field and distinguished from intracellular membranes assessed by whole-cell...

Mapping the local organization of cell membranes using excitation-polarization-resolved confocal fluorescence microscopy.

Fluorescence anisotropy and linear dichroism imaging have been widely used for imaging biomolecular orientational distributions in protein aggregates, fibrillar structures of cells, and cell membranes. However, these techniques do not give access to complete orientational order information in a whole image, because their use is limited to parts of the sample where the average orientation of mol...
